DESCRIPTION:Full-length,untaggedhumanrecombinanthistoneH1.0expressedinE.coli.H1histonesfunctionas‘linkers’thatbindtointernucleosomalDNA(~1:1withthehistoneoctamerofthenucleosomecore),convertingnucleosomesto“chromatosomes”andpromotingtheformationofmorecompact,higher-orderchromatinstructure.HistoneH1.0,theH1variantmosthomologoustotheH5histoneofavianerythrocytes,is,alongwithhistonesH1.1-H1.5,oneofsixH1variantsfoundinterminallydifferentiatedcells.Recentworkindicatesthatasidefromplayingaroleinchromatinstructureandfunction,histoneH1.0interactswithawidearrayofnucleolarproteins,withspecificinteractionsimplyingaroleinrRNAsplicingandribosomebiogenesis.Ingeneral,phosphorylationisthepredominantH1post-translationalmodification,notablyassociatedwithcellcycleprogression,butothermodifications,includinglysineacetylation/methylation1andADP-ribosylation10,arealsoreported.CrosstalkoccursbetweenH1modificationsandthoseonotherhistonesandH1variantscanaffectparticularhistonemodifications,e.g.H1.3(mouseH1d)inhibitionofSET7/9H3K4methylation(seedatasheet),consistentwitharoleforH1intranscriptionalregulation.Indeed,gene-specifictranscriptionalregulatoryeffectsofH1variantshavebeenreported,buttheseremainlesswellstudiedthanH1’sroleinchromatinstructure.
