DESCRIPTION:
HumanrecombinantSET8expressedinE.coliwithanN-terminalGSTfusion-tag.CatalyzesthetransferofmethylgroupsfromS-adenosyl-L-methionine(SAM)tothee-aminofunctionofproteinL-lysineresidues,specificallythemonomethylationoflysine-20ofhistoneH4(H4K20me1)andoflysine-382ofp53(p53K382me1).SET8andH4K20me1areessentialtochromosomecondensation,entryintomitosisandmaintenanceofgenomicstABIlityandareimplicatedinthelicensingofreplicationorigins.MonomethylationofH4K20andcellularSET8levelsoscillatewiththecellcycle,peakingattheG2/Mtransition,declininglateinmitosis,withSET8proteinbecomingnearlyundetectableinSphase.Apartfromitsroleincellcycleprogression,thereisevidenceSET8canregulatetranscriptionofspecificgenesbyH4K20monomethylationinpromoterelementsorgenebodiesandbymethylationofatranscriptionfactor,p53.MultiplelinesofevidencesuggestthatSET8maybeapromisingtargetforanti-cancertherapy.TheseincludetherequirementforSET8incellcycleprogression,SET8’sactivationofWnttargetgenes,itsnegativeregulationofthep53tumorsuppressor,theassociationofincreasedSET8levelswithbreastcancermetastasisandtheassociationofdecreasedlevelswithlongersurvivalinhepatocellularcarcinomapatients.
